He won’t be Nova, but Fillion will be in Guardians of the Galaxy as Cosmo… a talking dog.
Just the other day Guardians director James Gunn tweeted that Nathan Fillion will not be Nova in the upcoming film, but he did say that the actor would have a small cameo that was done ‘as a favor’ to the director.
Now, Schmoes is reporting that Fillion will be voicing Cosmo, the psychic Russian cosmonaut dog that can be found in Guardians comics. This is what they had to say;

Cosmo is a telepathic Russian dog in the Marvel Comics universe. The character was created by Dan Abnett and Andy Lanning, first appeared in Nova vol. 4 #7. Within the context of the stories, Cosmo is the security chief of the spacestation Knowhere and a member of the Guardians of the Galaxy.
So far, nothing has been confirmed from the production company.
